General Description of CD127 / IL7R / IL7RA.
The A7R34 antibody is specific for mouse CD127, a 60-90 kDa cell surface protein also known as the Interleukin-7 Receptor alpha chain, or IL-7R alpha. CD127 is typically expressed at the cell surface as a heterodimer with the common gamma chain (CD132). This complex acts as the functional receptor for IL-7, a cytokine important in T and B cell development, and in mature T cell homeostasis. A second cytokine known as Thymic Stromal Lymphopoietin (TSLP) also binds to a receptor complex of CD127 and the TSLPR chain to trigger activation of dendritic cells, and is involved in B cell development, allergy and autoimmunity.The A7R34 antibody may be used as a phenotypic marker for CD127 on immature B cells, on subsets of thymocytes which are double negative (CD4-CD8- ) or single positive (CD4+ or CD8+), and at low levels on mature, peripheral T cells. CD127 is a key marker, when used in combination with CD4 and CD25, to distinguish Treg and effector/memory Treg populations known as T(REM).

Staining with anti-CD127 / IL7R / IL7RA antibody reveals CD127 / IL7R / IL7RA expression is expected to be primarily localized to the cytosol and nucleoplasm.

Observed Antibody Staining Data By Tissue Type:

Variations in CD127 / IL7R / IL7RA antibody staining intensity in immunohistochemistry on tissue sections are present across different anatomical locations. An intense signal was observed in exocrine glandular cells in the pancreas, glandular cells in the thyroid gland and non-germinal center cells in the tonsil. More moderate antibody staining intensity was present in exocrine glandular cells in the pancreas, glandular cells in the thyroid gland and non-germinal center cells in the tonsil. Low, but measureable presence of CD127 / IL7R / IL7RA could be seen in cells in the tubules in kidney, epidermal cells in the skin, glandular cells in the endometrium, fallopian tube, gallbladder, prostate and stomach, hepatocytes in liver, Leydig cells in the testis, myocytes in skeletal muscle, neuronal cells in the caudate nucleus and cerebral cortex and squamous epithelial cells in the esophagus. We were unable to detect CD127 / IL7R / IL7RA in other tissues. Disease states, inflammation, and other physiological changes can have a substantial impact on antibody staining patterns. These measurements were all taken in tissues deemed normal or from patients without known disease.

Tissues from cancer patients, for instance, have their own distinct pattern of CD127 / IL7R / IL7RA expression as measured by anti-CD127 / IL7R / IL7RA antibody immunohistochemical staining.
