General Description of CD45R / B220.
The RA3-6B2 antibody reacts with the human and mouse CD45 isoform known as CD45R, or B220, a protein tyrosine phosphatase of 220 kDa. CD45 is one of the most abundant hematopoietic markers, and is expressed on all leukocytes (the Leukocyte Common Antigen, LCA). Various isoforms are generated and expressed in cell-specific patterns, all critical for leukocyte function. In mouse, the CD45R/B220 isoform is predominantly found on B cells, at varying levels on all stages from pro-B cells to activated B cells, and may also be detected on certain T cell and NK cell subsets. It is of note that B220 is not similarly expressed on human B cells, where it appears to be differentiation-specific and therefore expressed on only some B cell subsets. Other forms of CD45 with restricted cellular expression include CD45RA, CD45RB, CD45RO and several others.The RA3-6B2 antibody is one of the most consistently used leukocyte markers for B cells, T cell subsets and NK cell subsets in human and mouse. This antibody has also been reported as cross-reactive with feline CD45R/B220.

CD45R / B220 Previously Observed Antibody Staining Patterns

Observed Antibody Staining Data By Tissue Type:

Variations in CD45R / B220 antibody staining intensity in immunohistochemistry on tissue sections are present across different anatomical locations. An intense signal was observed in lymphoid tissue in appendix, hematopoietic cells in the bone marrow, germinal center cells in the lymph node, non-germinal center cells in the lymph node, cells in the red pulp in spleen, cells in the white pulp in spleen, germinal center cells in the tonsil and non-germinal center cells in the tonsil. More moderate antibody staining intensity was present in lymphoid tissue in appendix, hematopoietic cells in the bone marrow, germinal center cells in the lymph node, non-germinal center cells in the lymph node, cells in the red pulp in spleen, cells in the white pulp in spleen, germinal center cells in the tonsil and non-germinal center cells in the tonsil. Low, but measureable presence of CD45R / B220 could be seen in. We were unable to detect CD45R / B220 in other tissues. Disease states, inflammation, and other physiological changes can have a substantial impact on antibody staining patterns. These measurements were all taken in tissues deemed normal or from patients without known disease.

Tissues from cancer patients, for instance, have their own distinct pattern of CD45R / B220 expression as measured by anti-CD45R / B220 antibody immunohistochemical staining. The average level of expression by tumor is summarized in the table below. The variability row represents patient to patient variability in IHC staining.
