General Description of Laminin, gamma 1.
Laminins are large hetero-trimeric, non-collagenous glycoproteins composed of alpha, beta,, γ chains. This MAb reacts with laminin B2/1 chain of ~210kDa, does not cross-react with other basement membrane components or fibronectin. Its specificity was established by immunoprecipitation, immunofluorescence of human skeletal muscle, kidney with laminin chain-specific MAbs. Epithelial sheets in vivo are separated from the mesenchymal elements of the stroma by a thin layer of a specialized type of extracellular matrix termed the basement membrane (BM). This structure consists of individual components, some of which are ubiquitous in BMs, some are not. The ubiquitous ones comprise laminin (LN), entactin/niCanineen (EN), collagen type IV (CIV),, large heparan sulfate proteoglycan (HSPG), which interact specifically with each other to form a continuous, regular BM. Alterations of BM integrity, from local discontinuities up to complete loss, are described in many types of human, animal epithelial neoplasms. This MAb stains uniformly all human, murine basement membranes.

Anti-LAMC1 antibody staining is expected to be primarily localized to the endoplasmic reticulum and plasma membrane.

Observed Antibody Staining Data By Tissue Type:

Variations in Laminin, gamma 1 antibody staining intensity in immunohistochemistry on tissue sections are present across different anatomical locations. An intense signal was observed in trophoblastic cells in the placenta. More moderate antibody staining intensity was present in trophoblastic cells in the placenta. Low, but measureable presence of Laminin, gamma 1 could be seen inadipocytes in mesenchymal tissue, cells in the endometrial stroma in endometrium, cells in the seminiferous ducts in testis, cells in the white pulp in spleen, epidermal cells in the skin, fibroblasts in mesenchymal tissue, glandular cells in the appendix, colon, endometrium, gallbladder, prostate, rectum, salivary gland, stomach and thyroid gland, keratinocytes in skin, Leydig cells in the testis, neuronal cells in the caudate nucleus, ovarian stroma cells in the ovary, peripheral nerve in mesenchymal tissue, respiratory epithelial cells in the nasopharynx, squamous epithelial cells in the cervix and uterine, oral mucosa and vagina. We were unable to detect Laminin, gamma 1 in other tissues. Disease states, inflammation, and other physiological changes can have a substantial impact on antibody staining patterns. These measurements were all taken in tissues deemed normal or from patients without known disease.

Observed Antibody Staining Data By Tissue Disease Status:

Tissues from cancer patients, for instance, have their own distinct pattern of Laminin, gamma 1 expression as measured by anti-Laminin, gamma 1 antibody immunohistochemical staining. The average level of expression by tumor is summarized in the table below. The variability row represents patient to patient variability in IHC staining.
