General Description of Thyroglobulin.
Thyroglobulin is a 660kDa dimeric pre-protein with mutiple glycosylation sites. It is produced by, processed within the thyroid gland to produce the hormone thyroxine, triiodothyronine. Prior to forming dimers, thyroglobulin monomers undergo conformational maturation in the endoplasmic reticulation. The vast majority of follicular carcinomas of the thyroid will give positive immunoreactivity for anti-thyroglobulin even though sometimes only focally. Poorly differentiated carcinomas of the thyroid are frequently anti-thyroglobulin negative. Adenocarcinomas of other-than-thyroid origin do not react with this antibody. This antibody is useful in identification of thyroid carcinoma of the papillary, follicular types. Presence of thyroglobulin in metastatic lesions establishes the thyroid origin of tumor. Anti-thyroglobulin, combined with anti-calcitonin, can identify medullary carcinomas of the thyroid. Furthermore, anti-thyroglobulin, combined with anti-TTF1, can be a reliable marker to differentiate between primary thyroid, lung neoplasms.

Thyroglobulin Previously Observed Antibody Staining Patterns

Observed Antibody Staining Data By Tissue Type:

Variations in Thyroglobulin antibody staining intensity in immunohistochemistry on tissue sections are present across different anatomical locations. An intense signal was observed in glandular cells in the thyroid gland. More moderate antibody staining intensity was present in glandular cells in the thyroid gland. Low, but measureable presence of Thyroglobulin could be seen in. We were unable to detect Thyroglobulin in other tissues. Disease states, inflammation, and other physiological changes can have a substantial impact on antibody staining patterns. These measurements were all taken in tissues deemed normal or from patients without known disease.

Tissues from cancer patients, for instance, have their own distinct pattern of Thyroglobulin expression as measured by anti-Thyroglobulin antibody immunohistochemical staining. The average level of expression by tumor is summarized in the table below. The variability row represents patient to patient variability in IHC staining.
