Analysis of Protein Array containing more than 19,000 full-lengthhuman proteins using CDX2 Mouse Monoclonal Antibody (PCRP-CDX2-1A3) Z- and S- Score: The Z-score represents the strength of a signal that a monoclonal antibody (MAb) (in combination with a fluorescently-tagged anti-IgG secondary antibody) produces when binding to a particular protein on the HuProtTM array. Z-scores are described in units of standard deviations (SD’s) above the mean value of all signals generated on that array. If targets on HuProtTM are arranged in descending order of the Z-score, the S-score is the difference (also in units of SD’s) between the Z-score. S-score therefore represents the relative target specificity of a MAb to its intended target. A MAb is considered to specific to its intended target, if the MAb has an S-score of at least 2.5. For example, if a MAb binds to protein X with a Z-score of 43 and to protein Y with a Z-score of 14, then the S-score for the binding of that MAb to protein X is equal to 29.

General Description of CDX2 / Caudal Type Homeobox 2 (GI Epithelial Marker).
The specificity of this monoclonal antibody to its intended target was validated by HuProtTMArray, containing more than 19,000, full-length human proteins. The intestine-specific transcription factors CDX1 and CDX2 are important for directing intestinal development, differentiation, proliferation and maintenance of the intestinal phenotype. CDX2 protein expression has been seen in GI carcinomas. Anti-CDX2 has been useful to establish GI origin of metastatic adenocarcinomas and carcinoids and is especially useful to distinguish metastatic colorectal adenocarcinoma from lung adenocarcinoma. However, mucinous carcinomas of the ovary also express CDX2 protein. It limits the usefulness of this marker in the distinction of metastatic colorectal adenocarcinoma from mucinous carcinoma of the ovary.

CDX2 / Caudal Type Homeobox 2 (GI Epithelial Marker) Previously Observed Antibody Staining Patterns

Observed Subcellular, Organelle Specific Staining Data:

Anti-CDX2 antibody staining is expected to be primarily localized to the nucleoplasm. There is variability in either the signal strength or the localization of signal in nucleoplasm from cell to cell.

Observed Antibody Staining Data By Tissue Type:

Variations in CDX2 antibody staining intensity in immunohistochemistry on tissue sections are present across different anatomical locations. An intense signal was observed in glandular cells in the appendix, colon, duodenum, rectum. More moderate antibody staining intensity was present in glandular cells in the appendix, colon, duodenum, rectum. Low, but measureable presence of CDX2 could be seen in. We were unable to detect CDX2 in other tissues. Disease states, inflammation, and other physiological changes can have a substantial impact on antibody staining patterns. These measurements were all taken in tissues deemed normal or from patients without known disease.
