Antibody (Suitable for clinical applications)
Specification | Recommendation |
---|---|
Recommended Dilution (Conc) | 1:25-1:50 |
Pretreatment | EDTA Buffer pH 8.0 |
Incubation Parameters | 30 min at Room Temperature |
Prior to use, inspect vial for the presence of any precipitate or other unusual physical properties. These can indicate that the antibody has degraded and is no longer suitable for patient samples. Please run positive and negative controls simultaneously with all patient samples to account and control for errors in laboratory procedure. Use of methods or materials not recommended by enQuire Bio including change to dilution range and detection system should be routinely validated by the user.
Bcl-6 Oncoprotein Information for Pathologists
Summary:
Transcription factor at 3q27 normally expressed in germinal centers and in 5 – 15% of intrafollicular CD4+ T cells. Protects germinal center (GC) B cells against DNA damage induced apoptosis during somatic hypermutation and class switch recombination (J Exp Med 2010;207:1209). Translocations help define double hit lymphomas (Am J Clin Pathol 2010;134:258). Interpretation Nuclear staining.
Common Uses By Pathologists:
Germinal center marker (also CD10). Marker for follicular lymphoma (30 – 80%) of cases, can distinguish from MALT (BCL6-). Double staining of CD3 and BCL6 is helpful in appreciating follicular T helper cell immunophenotype of angioimmunoblastic T cell lymphoma (Hum Pathol 2010;41:79). Associated Diseases (KEGG IDs) | Chromosomal aberrations involving BCL6 are a cause of B-cell non-Hodgkin lymphomas (B-cell NHL), including diffuse large B-cell lymphoma and follicular lymphoma. Approximately 40% of diffuse large B-cell lymphomas and 5 to 10% of follicular lymphomas are associated with chromosomal translocations that deregulate expression of BCL6 by juxtaposing heterologous promoters to the BCL6 coding domain (PubMed:10469447, PubMed:10753856, PubMed:12414651, PubMed:11821949). General Description of Bcl-6 Oncoprotein . | |
This antibody reacts with the Bcl-6 gene product in follicular lymphomas, diffuse large B cell lymphomas, Burkitt’s lymphomas, and in nodular, lymphocyte predominant Hodgkin’s disease. Bcl-6 is a proto-oncogene that encodes Kruppel-type Zinc-finger protein of 95 kD and shares homology with transcription factor. Bcl-6 is mainly expressed in normal germinal center B cells and related lymphomas. |
