Antibody (Suitable for clinical applications)
Specification | Recommendation |
---|---|
Recommended Dilution (Conc) | 1:50-1:100 |
Pretreatment | EDTA Buffer pH 8.0 |
Incubation Parameters | 15 min at Room Temperature |
Prior to use, inspect vial for the presence of any precipitate or other unusual physical properties. These can indicate that the antibody has degraded and is no longer suitable for patient samples. Please run positive and negative controls simultaneously with all patient samples to account and control for errors in laboratory procedure. Use of methods or materials not recommended by enQuire Bio including change to dilution range and detection system should be routinely validated by the user.
p40 Information for Pathologists
Summary:
One of 10 p63 isoforms (J Virol 2007;81:11569), important to classify non small cell lung cancer into adenocarcinoma (p40-) versus squamous cell carcinoma (p40+), which determines chemotherapy regimen. Truncated, nontransactivating isoform highly specific for squamous / basal cells. Terminology Also called ?Np63, p40. Different from these p40s:.
Notable Clinical Features:
Mutations cause ectrodactyly-ectodermal dysplasia-cleft syndrome, ankyloblepharon-ectodermal dysplasia-clefting syndrome, nonsyndromic split-hand/split-foot malformation (J Cell Sci 2011;124:2200). Uses by pathologists Classify non-small cell lung carcinoma on small biopsies or cell blocks:. Adenocarcinoma (?Np63- or Squamous cell carcinoma (?Np63+ [strong] and >50% of cells, TTF1-). Adenosquamous (?Np63 strong and less than 50%, J Thorac Oncol 2012;7:281, Am J Surg Pathol 2012;36:895).
Common Uses By Pathologists:
Classify non-small cell lung carcinoma on small biopsies or cell blocks:. Adenocarcinoma (?Np63- or Squamous cell carcinoma (?Np63+ [strong] and >50% of cells, TTF1-). Adenosquamous (?Np63 strong and less than 50%, J Thorac Oncol 2012;7:281, Am J Surg Pathol 2012;36:895). May help classify large cell lung carcinoma as part of panel (Am J Clin Pathol 2011;136:773). General Description of p40 . | |
p40 is a relatively unknown antibody that recognizes ΔNp63-a p63 isoform suggested to be highly specific for squamous/basal cells. In a recent study, p40 is equivalent to p63 in sensitivity for squamous cell carcinoma, but it is markedly superior to p63 in specificity1, which eliminates a potential pitfall of misinterpreting a p63-positive adenocarcinoma or unsuspected lymphoma as squamous cell carcinoma. These findings strongly support the routine use of p40 in place of p63 for the diagnosis of pulmonary squamous cell carcinoma. |
