Staining with mouse monoclonal ZAP70 [Clone 2F3.2] antibody in formalin-fixed paraffin-embedded human tonsil.Analysis of Mass Spec data (dashed-line) of fractions stained with ZAP70 MS-QAVA™ monoclonal antibody [Clone: 2F3.2] (solid-line), reveals that less than 2.3% of signal is attributable to non-specific binding of anti-ZAP70 [Clone 2F3.2] to targets other than ZAP70 protein. Even frequently cited antibodies have much greater non-specific interactions, averaging over 30%. Data in image is from analysis in Jurkat, U202 and HeLa cells.

General Description of ZAP70.
ZAP70 is a 70kDa protein tyrosine kinase found in T-cells, natural killer cells. Control of this protein translation is via the IgVH gene.In Western Blot of whole cell lysates of normal peripheral blood mononuclear cells, the antibody labels a band corresponding to ZAP70. In Western Blot of whole cell lysates of CD19-positive purified leukemia cells from patients with Ig-unmutated, Ig-mutated CLL, the antibody labels a band corresponding to ZAP70 in the Ig-unmutated CLL samples, whereas no band is observed in the Ig-mutated CLL samples. In Western Blot of cell lysates of Jurkat cells (T-lymphoblastic cell line), the antibody labels a band of 70kDa protein. In Western Blot of cell lysates of A431 cells (carcinoma cell line), no band is observed. ZAP70 protein is expressed in leukemic cells of approximately 25% of chronic lymphocytic leukemia (CLL) cases as well. Anti-ZAP70 expression is an excellent surrogate marker for the distinction between the Ig-mutated (anti-ZAP70 negative), Ig-unmutated (anti-ZAP70 positive) CLL subtypes, can identify patient groups with divergent clinical courses. The anti-ZAP70 positive Ig-unmutated CLL cases have been shown to have a poorer prognosis.

ZAP70 Previously Observed Antibody Staining Patterns

Observed Subcellular, Organelle Specific Staining Data:

Variations in ZAP70 antibody staining intensity in immunohistochemistry on tissue sections are present across different anatomical locations. An intense signal was observed in lymphoid tissue in appendix and non-germinal center cells in the lymph node and tonsil. More moderate antibody staining intensity was present in lymphoid tissue in appendix and non-germinal center cells in the lymph node and tonsil. Low, but measureable presence of ZAP70 could be seen inmacrophages in lung and cells in the white pulp in spleen. We were unable to detect ZAP70 in other tissues. Disease states, inflammation, and other physiological changes can have a substantial impact on antibody staining patterns. These measurements were all taken in tissues deemed normal or from patients without known disease.

Tissues from cancer patients, for instance, have their own distinct pattern of ZAP70 expression as measured by anti-ZAP70 antibody immunohistochemical staining. The average level of expression by tumor is summarized in the table below. The variability row represents patient to patient variability in IHC staining.
