MS QAVA™:  Quantitative Measurement of Antibody Specificity Powered by Mass Spec.

Existing validation only provides approximations and assumptions of specificity. Here’s how it works:

Analysis of Mass Spec data (dashed-line) of fractions stained with EGFR (Her1 / ErbB1) QAVA mAb [Clone H9B4] (solid line) reveals that less than 10% of signal is attributable to non-specific binding of H9B4 to targets other than EGFR (Her1 / ErbB1). This specificity is excellent compared to the average antibody which shows as much as 30% non-specific signal. Data from Jurkat, U2oS and HeLa cells.

Immunoprecipitation and Mass Spec is run for every fraction for a total of 36-72 sample types per antibody. Specificity is calculated as antibody vs. mass spec r-squared.  r-squared represents variability in antibody signal attributable to variability in level of protein (as measured by mass spec).
