General Description of Desmocollin-2/3.
Desmosomes are intercellular adhering junctions that represent cell surface attachment sites for intermediate filament. The desmosome is subdivided into two regions. The plaque region lies adjacent to the plasma, and is believed to contain molecules that attach the intermediate filament cytoskeleton to the desmosome. The core region is composed of transmembrane glycoproteins that are thought to mediate cell-cell adhesion. Desmogleins and desmocollins are the main desmosomal transmembrane proteins. These desmosomal glycoproteins belong to the members of the cadherin family of adhesion molecules. Three different isoforms of both desmogleins and desmocollins have been identified, named as desmoglein 1-3 and desmocollins. Desmosomal cadherins showed differentiation-specific expression in the human epidermis, although the functional significance of this differential expression is not fully understood. Desmocollin-1 can be found in the upper layers. The expression of desmocollin-2 varies in the basal and suprabasal layers. And desmocollin-3 is expressed more evenly throughout the suprabasal layers.

Observed Antibody Staining Data By Tissue Type:

Variations in Desmocollin-2/3 antibody staining intensity in immunohistochemistry on tissue sections are present across different anatomical locations. An intense signal was observed in epidermal cells in the skin, glandular cells in the appendix, endometrium, rectum and small intestine, myocytes in heart muscle and squamous epithelial cells in the esophagus, oral mucosa, tonsil and vagina. More moderate antibody staining intensity was present in epidermal cells in the skin, glandular cells in the appendix, endometrium, rectum and small intestine, myocytes in heart muscle and squamous epithelial cells in the esophagus, oral mucosa, tonsil and vagina. Low, but measureable presence of Desmocollin-2/3 could be seen inglandular cells in the endometrium, epididymis, gallbladder, seminal vesicle and thyroid gland, hepatocytes in liver, keratinocytes in skin, macrophages in lung, respiratory epithelial cells in the bronchus, urothelial cells in the urinary bladder and glandular cells in the endometrium, epididymis, gallbladder and seminal vesicle and thyroid gland. We were unable to detect Desmocollin-2/3 in other tissues. Disease states, inflammation, and other physiological changes can have a substantial impact on antibody staining patterns. These measurements were all taken in tissues deemed normal or from patients without known disease.

Tissues from cancer patients, for instance, have their own distinct pattern of Desmocollin-2/3 expression as measured by anti-Desmocollin-2/3 antibody immunohistochemical staining. The average level of expression by tumor is summarized in the table below. The variability row represents patient to patient variability in IHC staining.
